ヘルペスワクチン最前線 15: Tomegavax

では、早速、「ワクチンリスト解説 第15弾！」を始めたいと思います…。

今回は、TomegaVax というワクチンで、アメリカ オレゴン州ポートランドにある Oregon Health & Science University （OHSU）からの Spin Off で始まった TomegaVax.Inc という会社（＝大学での研究から派生した会社）が研究・開発を進めているものであります。

ポートランドといえば、Starbucks 発祥の地ですからね！今回のオープニング画像は、それにちなんで Starbucks にしてみました (＾◇＾)




（１）TomegaVax ってどんな会社なの？

まぁ、いつものように「TomegaVax のサイトを探してみよう！」と思いながら、インターネットをウロウロしていたのですが、なかなか見つからず「何でなかなか見つからないのだろうと」不思議に思っていました。

Google で検索してみると、

　　「TomeVax Acquired…」（TomeVax が買収…）

といった記事がちらほらと出てきたので、ちょっとビックリ＆慌てながらも、いろいろと情報を調べてみました。

まずは、「会社を始めるには資金が必要なわけだから、おそらく、 以前少しだけ紹介した SBIR（Small Business Innovation Research「中小企業技術革新制度」）などを使って資金調達をするのではないか」という予測の基に SBIRのホームページから情報を探してみました。

すると、

　　バッチリ、ありました！(＾◇＾)

探し当てた情報が以下のものです！

申請及び資金の受諾が、今から3年前の2014年、SBIRから提供された資金が299,863ドル。加えて、資金提供が複数年度ではなく、2014年度のみとなっていることを考えれば、

　　おそらくもうすでに SBIRから提供された資金は底をついている

と想像するのが普通でしょう。だとすれば、先程少し紹介した買収の話もあながち嘘ではないということになるわけです…。

そこで、この買収の件についてもう少し調べてみることにしました。

（２）TomegaVax 買収の件について

Google で検索をしていると、TomegaVax Acquired と書いてある記事がいくつか出てくるので、そのうちの一つをクリックしてみたところ、以下のような OHSU 内のサイトに出くわしたんです。

やけに先生がニコニコしているので、

　　「買われたっていうのに、何をニコニコしとるんじゃい！」( ﾟДﾟ)

と思ってしまったのですが、もう少し詳しい情報が書いてある記事を読むと、結構いい感じでの買収だったようなんです。その記事というのが、以下のものになります。

A San Francisco-based biotechnology company that publicly launched today has acquired Portland-based TomegaVax Inc., an Oregon Health & Science University spinoff that is pursuing an HIV vaccine.

Vir Biotechnology is taking on “some of the world’s most challenging infectious diseases for which solutions are non-existent or inadequate,” the company’s announcement says. The company is "seeking to apply immune programming on an unprecedented scale."

TomegaVax will continue to conduct its research in Portland, despite its acquisition by a San Francisco-based startup.

“Vir is adopting a broad technological portfolio, including the viral vectors obtained through the acquisition of TomegaVax,” it announced.

Those technologies originated at the OHSU Vaccine & Gene Therapy Institute, led by Dr. Louis Picker and Klaus Frueh. Picker serves as one of Vir’s scientific advisors and Frueh as a director.

“This is a huge step for us,” Frueh said. “Our vaccine development work at OHSU has now matured to the point where we can start clinical testing in people. It’s a completely different ballgame from the academic research we’ve been doing. Vir is going to become our commercial development partner to move the vaccines to people.”

ARCH Venture Partners and the Bill & Melinda Gates Foundation are among the lead investors. ARCH Venture, whose co-founder Robert Nelson led the formation of Vir, has committed to invest $150 million.

While TomegaVax is now a wholly owned subsidiary of Vir, it will continue to be based at the OTRADI Bioscience Incubator. The company was able to double its staff to 10 employees after the acquisition last September by what became Vir.

“The great news is that the VC funders were very impressed with the research and incubation facilities here in Oregon and are keeping the TomegaVax R&D team intact and headquartered here at the OTRADI Bioscience Incubator,” said Jennifer Fox, the Incubator’s executive director. “This is probably the biggest and best news we’ve had since opening our doors.”

Founded in 2011, TomegaVax is developing cures for AIDS, malaria, tuberculosis, herpes, papillomavirus and hepatitis — diseases where there’s no natural immunity in humans and traditional vaccine mechanisms don’t work. The approach is to use the CMV virus to trigger a persistent immune response and redirect it to other pathogens.

“That is really unique and has a lot of potential in many different applications and diseases because you can basically alert the immune system in a new way so you have an immune response to combat pathogens that were difficult to combat in the past,” Frueh said.

TomegaVax had been operating on small business grants and an alliance with a large pharmaceutical company. Financial terms of the deal with Vir were not disclosed.

気になる箇所に下線を引いてみたんですが、その内容をまとめてみると…、

• サンフランシスコのVir Biotechnologyという会社がTomegaVa.incを買収する模様
• Virへの主な出資者は、ARCH Venture Partners と Bill & Melinda Gates Foundation
• Virに買収された後、TomegaVaxは今まで通り、ポートランドで研究・開発を行う模様
• Virに買収してもらったおかげで、研究員も10名に倍増することができた

ということのようです。

まぁ、買い手となった Vir の後ろには、あの天下の「ビルゲーツ財団」がいるわけですからね…、そりゃぁ、良い買収になったと思います！研究員を10名に倍増することが出来たという点も、当事者にとってはとてもプラスになったと思いますね☆

総じて、

　　TomegaVaxちゃん、良いお家にもらわれて、良かったね♡

という感じの買収劇になるかと思います☆

（３）で、TomegaVax ってどうなの？今後はどうなるの？

会社としての TomegaVax については、ここまで書いたように色々と調べることが出来たのですが、肝心の TomegaVax 自身については、ほとんど情報が見当たらなかったんです… (´･ω･｀)

唯一見つかったものとしては、先ほどの SBIR への申請書でして…、それ以外はほとんど記載が見られませんでした…。そこで、SBIRへの申請書に書いてあることを見ると…、

DESCRIPTION (provided by applicant): The longterm goal of this project is to utilize cytomegalovirus (CMV) vectors in the development of a therapeutic vaccine against herpes simplex virus 2 (HSV-2), the causative agent of genital herpes. HSV-2 is widely prevalent in the population causing recurrent genital lesions and significantly increasing the risk for other sexually transmitted diseases such as HIV. Despite this unmet medical need, there is no approved vaccine for HSV-2. While some vaccine approaches have shown promise in preclinical studies, they failed to protect individuals from de novo or recurrent infection in clincal trials. Given the known role of T lymphocytes in limiting HSV-2 reactivation and the associated development of lesions, a successful HSV-2 vaccine needs to induce and maintain a strong T cell response that effectively identifies and neutralizes infected cells. CMV vectors are uniquely qualified for this task due to their ability to induce lifelong, high frequency effector memory T

というものらしく、 技術的には「サイトメガロウィルスのをベクター（宿主）としてワクチンを作成する」ことがカギになっているようで、他のウィルス（HIVなど）へのワクチン製造にも同じ技術を使っているようです。

【参考までに…】
サイトメガロウィルスは、いわば「口唇・性器ヘルペスの大元締め」に相当するもので、帯状発疹や乳幼児突発性発疹（←これは誰もが感染するものです）もこの中の一つになります。「横浜市衛生研究所」によると、以下のような説明がなされています。

「サイトメガロウィルス感染症」について 

サイトメガロウイルスは、ヒトヘルペスウイルス(HHV)の仲間です。ヒトヘルペスウイルスには、1型から8型まであります。サイトメガロウイルスは、ヒトヘルペスウイルスの5型（HHV-5）にあたります。ヒトヘルペスウイルス(HHV)の仲間を列挙してみると、以下のとおりです。

HHV-1 :　単純ヘルペスウイルス1型(HSV-1)。口唇ヘルペスの病原体です。
HHV-2 :　単純ヘルペスウイルス2型(HSV-2)。性器ヘルペスの病原体です。
HHV-3 :　水痘帯状疱疹ウイルス(VZV)。水痘と帯状疱疹の病原体です。
HHV-4 :　Epstein-Barrウイルス(EBV)。伝染性単核症の病原体です。
HHV-5 :　サイトメガロウイルス(CMV)。
HHV-6 :　突発性発疹の病原体です。
HHV-7 :　突発性発疹の病原体です。HHV-6より発生は少ないです。
HHV-8 :　同性愛の男性で感染している割合が高く、男性間の性行為で感染し、ＨＩＶ感染者でのカポジ肉腫の発生に関与していると考えられています。

これらのヒトヘルペスウイルス(HHV)は、初めての感染を受け症状が消えた後も、長期にわたって体内に潜伏・休眠状態で留まることができるという特徴を持っています。そして、ウイルスによっては病気や薬によって免疫がひどく低下したときにウイルスが潜伏・休眠状態から再び活性化し何らかの症状を現すことがあります。

では、TomegaVax の説明に戻ることにしましょう…！

ここで、注意してもらいたいのは…、

このメガロウィルスをベクター細胞としてワクチンを作るという技術は、HIVやB型肝炎などの感染症に対処するために開発したものであって、何も口唇・性器ヘルペスワクチンを作るために開発したものではない

ということです…。

研究に資金が必要なのは当然のことなのですが、その資金を引き出すには、人類にとってより大きな脅威となっている感染症であることが大事です。「あまり重要でない疾病にお金をかける必要はない…」という論理がそこにはあるのでしょうが…、今回の買収に関しても、

Vir側としては、TomegaVa.incが持っているワクチン開発技術を活用して、B型肝炎などのより重篤な感染症に対応するワクチン開発を行うために買収した可能性が高い 

と考えられます。

事実、CISIONという科学技術＆企業系のサイトでは、今回の買収に関して以下のような記事が掲載されておりました。

Chronic and acute infections impact hundreds of millions of people every year. For example, according to the World Health Organization, there are about 240 million people in the world living with chronic Hepatitis B, with millions of new infections annually. Tuberculosis is one of the leading causes of death globally—two billion have latent infection and ten million new cases occur annually. このような状況から見ると…、

この Tomegavax に関しては、可能性が絶対100％ゼロというわけではないだろうけど…、性器ヘルペスのワクチン開発に転用される可能性はあまり高いとは言えないだろう…。

 ということになるかと思います… 涙。

という考えが浮かんできました！この「TomegaVaxがHSVにスッゴク効く！」というのであればやる価値はあるでしょうが、それも分からないのでは…、う～ん、なかなか難しいかもしれないですが、故 Halford博士の TheraVax であれば、結構イケルかもしれないですよね♪
