- Agenus社: HerpV （治療目的）
既感染者に対する治療を目的としたワクチンで、2012年中頃には治験を開始する予定。Phase Iでは、HSVウイルスに対抗するCD+4 T細胞ならびにCD+8T細胞の反応が活性化されたようで、非常に良い結果であった模様…。
AgenusのCEOであるGaro H. Armenは、Phase IIは開始後1年半ほどかかるようで、「少しだけ早くなる」可能性もあるようです。
- Genocea & Isconova （治療＆予防目的）
Genoceaというバイオ系の会社ですが、最近Isoconova製のMatrix Mという免疫活性剤（？）を利用する契約を取り付けたようで、HVS2を含む6つの感染症に対する効果の検証を行っているようです。
現在、その中で最も進んでいる研究は、HSV2をターゲットとしたもので、2012年には人間を対象とした治験を開始する予定のようです。
GenoceaのCEO（Chip Clark）によると、「T細胞やB細胞を活性化させる安全で効果的な活性剤は、（HSV２という）世界中に広がっている感染症をこれ以上拡散させないためには非常に大切なものである。この目的に資するために、Isconovaと手を組み、Matrix Mを使用するのは、非常に喜ばしいことと考えている」ようです！
- Vical （治療目的）
VicalのCEOを務めるVijay Samantによると、Vicalは「現在は、治験前の段階を完了し、人間を対象とした治験を開始するための準備を行なっているところであるが、同薬品の開発はVical内でも優先順位として非常に高いものである」という状況のようです。
臨床治験前段階での結果は以下の通り… - 糖タンパク質D+（もしくはD-）と二つのヘルペス外皮タンパク質を読み込ませた予防用ワクチンは、初回の感染だけでなく、既感染者の再発も完全に防ぐことができた。
- 同予防用ワクチンにより、非活性化状態にあるウイルスだけでなく、初回感染場所におけるウイルスの複製も少なくすることができた。
- 糖タンパク質D+と二つのヘルペス外皮タンパク質を読み込ませた治療用ワクチンは、HSVにすでに感染している動物において、再発症を優位に少なくすることができた。
- 同治療用ワクチンは、ウイルス感染部からの排出量を少なくすることができた。
- Sanofi Pasteur （不明）
ACAM-529は、不完全なヘルペスウイルスを複製したミュータント ウイルスであり、HSV1および2に対して効果があることが確認されている（人間ではまだ未確認…）。会社の代表となっているJim Tartagliaによると、「ACAM-529はまだ開発段階であり、2012年には治験Phase Iに入る予定」となっているようです。
- Coridon （治療＆予防目的）
2011年10月末、Allied Healthcare Groupから追加の研究資金を受けることが決定しており、治験Phase Iに入るまでに基本的安全性を証明するための治験前テストを行うように予定されているようです。
研究チームを引っ張っているFrazer教授は、「私たちが開発したヘルペスウイルスワクチンの結果は、エキサイティングな時代の幕開けとなると考えられ、次の12か月後には私たちのHSVワクチンが素晴らしい結果をもたらしてくれることになるでしょう」と述べているようです。おそらく、2013年までには地検が開始されていることでしょう…。
- Amgen - Immunovex （不明）
AmgenのImmunoVexですが、「Amgenが開発を中止したのではないのか…」、「2012年の2月か3月までにはPhase IIの治験が開始されるのでは…」というように色々とうわさされているようですが…。

の模様でして、「会社の方としても、今後Immunovexをどうするのか、方針も決まっていない」ということのようです…。
まぁ、つまりは、「Phase Iの結果次第」、ということでしょうか…

Here is a list of drugs and vaccines going (or advancing) into Human Clinical Trials in 2012:
- Agenus: HerpV
Therapeutic -- Agenus has stated that they plan to move their herpes vaccine, HerpV, into clinical trials around mid year in 2012.
Their results from Phase I: All patients who were evaluable for immune response and received HerpV showed a statistically significant CD4+ T cell response [used to search and hunt down pathogens] (100% 7/7) to HSV-2 antigens and the majority of those patients demonstrated a CD8+ T cell response [used to kill or destroy the pathogens] (75%; 6/8 ) .
Garo H. Armen, Chairman and CEO of Agenus, stated that the Phase II will last 18 months from the day the trial starts, a "conservative estimate" or "it may be sooner." Let's hope for sooner. - Genocea & Isconova: Therapeutic & Preventive
Genocea currently has the right to use Isconova's Matrix M in vaccines targeting six infectious diseases including programs in its pipeline for herpes simplex virus type-2, Chlamydia trachomatis, and Streptococcus pneumoniae.
The lead program of the collaboration is a therapeutic vaccine for herpes simplex virus type-2. Genocea expects to initiate human trials with this program in 2012.
Chip Clark, CEO of Genocea commented, “A safe and effective adjuvant that elicits strong B and T cell immune response is essential to our aim to prevent or treat the world’s most intractable infectious diseases. For that reason we are delighted to expand our collaboration with Isconova for Matrix M.” - Vical: Therapeutic
According to Vijay Samant, Vical is planning "to complete preclinical development and prepare our plan to advance the therapeutic vaccine into human clinical testing. The program is high on our priority list and we look forward to defining a path forward in the near future."
Their results in pre-clinical testing: Prophylactic vaccines encoding glycoprotein D plus or minus, two herpes virus tegument proteins provided complete protection from both primary and recurrent disease; the prophylactic vaccines also significantly reduced viral replication at primary infection site as well as detectable virus at the latent site; therapeutic vaccines encoding glycoprotein D plus, the two tegument protein significantly reduce the frequency of general lesion outbreaks in animals with pre-established HSV to infections; the therapeutic vaccines also significantly reduced the frequency of general viral shedding. - Sanofi Pasteur
ACAM-529 -- unsure of treatment type, Therapeutic and/or Preventive -- ACAM-529 is a replication-defective mutant virus that has proved successful both in preventing HSV-1/HSV-2 infections, and in combating the virus in already infected hosts, as in animal models, unsure of status in humans. According to Jim Tartaglia, a company representative of Sanofi Pasteur, ACAM-529 is still under development and should enter Phase I human clinical trials in 2012. - Coridon
Therapeutic and Preventive -- I had to add Coridon to the list or I wouldn't hear the end of it; even though they still have to do more preclinical (safety) testing for their herpes vaccine before they enter human clinical trials.
As of late October 2011, Coridon has now secured additional funding from major investor Allied Healthcare Group to begin manufacturing the vaccine and they have to conduct pre-clinical safety studies before testing the vaccine in a human Phase I clinical study.
“The results of our herpes vaccine mark the beginning of an exciting period," said Professor Frazer. "Over the next 12 months, we expect pivotal data showing that our HSV vaccine, which incorporates Coridon optimisation technology, produces similar immune responses in the clinic to those seen in the animal trials.” Which makes me think Coridon won't enter human clinical trials until 2013, but I could be wrong. - Amgen - Immunovex
Status Unknown, the drug may be scrapped or it may enter Phase II clinical trials by February or March 2012. Time will tell.
